The Bioenergetics group at the Techinical University of Berlin offer two master thesis / internship positions for motivated candidates on two interesting projects in Biotechnology. 

Master Thesis/Internship: Biosensors for applications in live cells.
Project: Probing a specific metabolite in Ralstonia Eutropha using a genetically encoded fluorescent biosensor.

Background: Genetically-encoded fluorescent sensors are the complex chimeric proteins consisting of a fluorescent protein and a sensing polypeptide fused in a specific manner. These sensors demonstrate clearly detectable changes in fluorescence in response to corresponding cell events. They also allow precise targeting into various subcellular compartments or animal tissues and aid in real time in vivo measurement of signals in natural intracellular surroundings. Biosensors monitoring changes in intracellular concentration of calcium ions (Ca2+),  hydrogen peroxide (H2O2), and Nicotinamide adenine dinucleotide (NADH) are successful examples of such kind.         
                  
Proposed Project: Ralstonia Eutropha contain hydrogenases which are fascinating redox proteins and biomolecule based electrocatalyst, showing tremendous promise in hydrogen production. Their hydrogenases play critical roles in both biohydrogen production and hydrogen oxidation during growth of R. Eutropha. In this project we propose to use a genetically encoded fluorescent protein based biosensor  developed for a biomolecule, providing clues about cellular metabolism and state. We are interested to test the ability of this biosensor to report different concentration of this biomolecule during different metabolic states in R. Eutropha. Detailed knowledge of metabolism in R. Eutropha is very essential for exploring it further in Biotechnology and Biofuels.

Methods: DNA cloning, Protein Expression in E. Coli and R. Eutropha,Protein Purification and Activity assay, Fluorescence microscopy and Imaging.
