“Kinetic Characterization of an in-vitro Enzymatic Cascade for Nucleotide Sugar Production by state-of-the-art Ion Exchange Chromatography”

Glycosylation is an important modification of proteins, and is involved in the regulation of biological activity and stability of many proteins. Notably, the glycosylation of many eukaryotic proteins appears to be highly-regulated and minor structural changes of glycan portions may lead to enzyme dysfunction. Research into glycosylation has, thus, profoundly increased over the last years and controlling protein glycosylation is seen as a key for the cost-effective manufacturing of biotechnological products. In a joint project with the University of Halle-Wittenberg, an in-vitro enzyme cascade consisting of endoplasmatic reticulum glycosyltransferases is build up to produce Nglycans enzymatically. Glycosyltransferases require nucleotide sugar-donors (e.g. GDP-mannose), which are highly priced chemicals that need to be produced enzymatically as well for an economically efficient production process. Thus, the proposed master project will focus on the kinetic characterization and optimisation of the enzymatic GDP-mannose production. The kinetic constants are determined by ion exchange chromatography.

- Background in biotechnology, bioengineering, biosystems technology, process engineering, biochemistry, analytical chemistry or related fields
- Ideally, experience with ion exchange chromatography and enzyme kinetics
- MS Office, ability to work in a team
