In the field of “Genomic views of protein-RNA interactions and splicing regulation” the research group of Dr Julian König offers the following PhD project:
Illuminating a key player in splicing regulation
The human RNA-binding protein PTB is a central regulator of alternative splicing with strong impact on neuronal differentiation. It recognizes a multitude of target exons to affect their splicing in distinct ways, leading to the coordinated expression of tissue-defining transcript isoforms. However, it is unclear how this versatile protein differentiates between activities and achieves specificity in activating or repressing certain exons.
The aim of this project is to dissect the PTB splicing regulatory network and its molecular mechanisms using next-generation sequencing-based technologies that are currently revolutionizing RNA biology. Among these, the PhD student will map protein-RNA interactions on a genome-wide scale using iCLIP and identify the responsible cis-regulatory splicing signals through a systematic mutational screen that we recently developed in the lab. These approaches will be complemented by the identification of protein partners using quantitative mass spectrometry. Insights from the genome-wide approaches will be followed up with genetic and biochemical experiments to nail down mechanistic principles of regulation.
