In the field of “Innate Lymphoid Cells” the research group of Prof. Andreas Diefenbach offers the following PhD project: Role of the diverse tid splice forms in Wnt/Wg mediated colon tumorigenesis and progression

Description:

The tid gene encodes three transcripts (long: L, intermediate: I und short: S) translated into three cytosolic (Tid-LC, Tid-IC,Tid-SC) and three mitochondrial proteins (Tid-Lm, Tid-Im, Tid-Sm). Interestingly, the cytosolic Tid proteins L and I have been found to interact simultaneously with diverse physiological partners, among them the adenomatous polyposis tumor suppressor (APC), component of the Wnt/Wg signaling causal in normal development and neoplastic transformation of colon and many other cancers. We have shown that loss of tid correlates with APC deregulation and colon cancer progression. The question whether the action of the distinct Tid proteins is independent, synergistic or contradictory in the proliferation versus differentiation of colon epithelia resulting in the cancerous growth has to be answered. To elucidate this topic in vivo splice form specific transgenic mtid-knock-in and -KO lines have been generated. With the help of these lines the developmental and tumorigenic impact of the single Tid forms in the neoplastic transformation and progression of colon epithelia will be investigated.
