PhD Position in Life Sciences

The International Research Training Group – Complex Membrane Proteins in Cellular Development and Disease (IRTG 1830) - jointly run by the University of Kaiserslautern (Germany), Saarland University (Germany) and the University of Alberta (Canada) - offers integrative approaches to biomedical research with a strong emphasis on developmental processes and diseases. The program is funded by the German Research Foundation (DFG) and the Natural Sciences and Engineering Research Council of Canada (NSERC).
Within the IRTG 1830, the following PhD Position is available in the lab of Prof. Dr. Manfred Schmitt (Molecular and Cell Biology, Dept. of Biosciences, Saarland University, Saarbrücken):

Yeast overexpression screen for cellular components restoring
plasma membrane trafficking of human kAE1
We use yeast as experimental system to identify gene products that affect the intracellular trafficking and turnover of a membrane protein (kAE1) that is vital for proper kidney function. Human kAE1 is a bicarbonate transporter in the basolateral membrane of renal epithelial cells that participates in the fine-tuning of acid-base homeostasis by mediating electroneutral Cl-/HCO3- exchange. Several autosomal mutations in the kAE1 encoding gene (SLC4A1) can cause clinical disorders known as distal renal tubular acidosis (dRTA) which are linked to kAE1 misfolding, ER/Golgi retention, and/or premature degradation. Since wild-type kAE1 can be expressed in yeast and is correctly targeted to the plasma membrane, the project will screen a yeast overexpression library for gene products that, when overexpressed, are capable to increase plasma membrane localization and restore transport activity of clinically relevant kAE1 trafficking mutants. The experimental approach will equally include aspects of yeast genetics, cell biology, protein biochemistry and physiology.

The program includes a 6-months stay in a lab of our Canadian partners and offers excellent scientific and transferable skills training at all participating locations. The program language is English and there are no tuition fees. The successful candidate will receive a contract (65% TVL13), as well as support with administrative matters, accommodation, visas etc. We invite applications from highly qualified and motivated students of any nationality and we are looking forward to your application for the PhD fellowship in our group.
