“New labeling strategies for quantitative single molecule imaging of membrane proteins”

 
The complex rules that govern the assembly of neurotransmitter receptors in synapses provide the molecular basis for the diverse functional repertoire of nerve cells. We recently developed a single molecule based imaging approach to decipher the composition of these receptors with unmatched precision. However, the availability of suitable fluorescent protein markers currently restricts the number of useable colors to two. The goal of this project is to develop novel approaches to utilize the potential of organic dye markers for imaging three or more colors simultaneously. The newly established labeling strategies will not only be applied to resolve the composition of membrane proteins, but also for other single molecule methods like superresolution imaging.

The work in the Ulbrich laboratory includes wet lab biology (cloning, biochemistry, cell culture), instrumentation (assembly of custom single-molecule microscopes), and programming to control equipment and evaluate results. Ideally, applicants should bring enthusiasm for science, creativity, technical and programming skills, as well as experience in microscopy, molecular biology and/or biochemistry. Ph. D. candidates should be independent and motivated individuals who hold a Diploma or M.Sc. degree in Biology, Physics, Chemistry or a related discipline.

The project is initially funded for 3 years by the German Research Foundation DFG but can be extended if required.
