In the field of “Deciphering DNA damage signaling using proteomics” the research group of Dr Petra Beli offers the following PhD project:
Understanding ubiquitin signaling in the cellular response to DNA damage
Posttranslational modification of proteins by ubiquitin is an essential mechanism regulating many cellular processes; in particular protein degradation and DNA repair. Ubiquitin ligases catalyze the attachment of the ubiquitin molecules to lysine residues in target proteins and thereby provide the specificity to the ubiquitin system. Mutations in a number of ubiquitin ligases are associated with neurodegenerative diseases or predisposition to the development of malignancies. However, the protein targets and functions of most ubiquitin ligases in human cells remain elusive. We have recently developed mass spectrometric methods to investigate cellular ubiquitylation patterns and employed these methods to study the response of the ubiquitin system to cellular perturbations including proteasome inhibition and genotoxic stress. The aim of this project is to identify protein substrates and functions of selected ubiquitin ligases implicated in DNA damage repair and signaling. To this end, the PhD candidate will employ a range of methods including mass spectrometry-based proteomics, cell biological and biochemical methods (flow cytometry, confocal microscopy, immunoprecipitation) and functional genomics.
