In the field of “Repair of structurally defined DNA lesions” the research group of Dr Andriy Khobta offers the following PhD project:
Interactions of DNA modifications and DNA repair with gene transcription
An innovative experimental system, which exploits ad hoc reporter vectors containing structurally defined DNA base and/or backbone modifications of interest in precisely defined positions, will be used to investigate the modes of interaction of bulky lesions, such as cyclobutane pyrimidine dimers and adducts of chemical carcinogens, with the RNA polymerase II transcription initiation and elongation machineries. The aim is to understand how the structure of DNA lesions and their localisation with respect to functional gene elements define the efficiencies of repair and how important are these parameters for the repair pathway choice. The project tasks will be (i) detection and investigation of the fate of stalled RNA polymerase complexes; (ii) determination of the efficiencies of repair of structurally different DNA adducts; (iii) identification of specific NER components required for the removal of different DNA adducts and for the recovery of transcription. 

The postgraduate student will obtain training in molecular cloning, transfection of mammalian cells, gene expression analyses, flow cytometry, chromatin immunoprecipitation and RNA techniques. Previous experience in any of the listed techniques would be advantageous but is not strictly required. However, applicants should demonstrate knowledge of and adherence to general laboratory rules as well as sound understanding of the research topic..
