Development of Muscle Spindles in Wildtype and Mutant Mice

Scientific background of the project: Muscle spindles are the main proprioceptors, i.e. they inform the brain about the extent and speed of muscle contraction and by this about the current positon of our extremities in space. They consist of specialized muscle fibers, called intrafusal fibers. Intrafusal fibers have a complex neuronal innervation pattern. Muscle stretch is sensed by the Ia afferent neuron that innervates the central part of intrafusal fibers, and the sensitivity to stretch is regulated and maintained by the g-motoneuron efferent innervation in both peripheral regions of intrafusal fibers.
We are seeking a PhD student to join our team working on the development and function of the neuronal innervation of muscle spindles. The project will be a continuation of previous studies (Zhang et al., 2014a Formation of cholinergic synapse-like specializations at developing murine muscle spindles. Developmental Biology 393: 227-235; Zhang et al., 2014b Differential Regulation of AChR Clustering in the Polar and Equatorial Region of Murine Muscle Spindles. Methodology: The project will involve confocal microscopy, transgenic mouse strains, electrophysiology, molecular biology and cell culture techniques.
