In the field of  “Chromatin-based mechanisms of genome maintenance” the research group of Dr. Vassilis Roukos offers the following PhD project:

Chromatin-based mechanisms of translocation biogenesis


Chromosome translocations are formed in the context of chromatin by the illegitimate joining of double-strand breaks (DSBs) to produce aberrant fusions. While translocations have been extensively characterized and their relevance in cancer is well established, how they form in the context of the intact cell nucleus is remarkably poorly understood. We have recently developed a unique cell-based system to follow the fate of individual DSBs as they form translocations in living cells and used it to analyze the steps leading to the formation of aberrant fusions (Roukos et al., Science, 2013). This established system has been used in pilot screens to identify chromatin modifiers that modulate translocation frequency. The aim of this PhD project is to elucidate how these identified chromatin modifiers contribute to translocations. The student will use the established translocation system in combination with state of the art live cell microscopy, cell biological methods and gain- or loss-of function experiments to characterize the effect of chromatin environment in the various steps of translocation biogenesis.
