In the field of  “Crosstalk between DNA damage checkpoints & nutrient signalling” the research group of Dr. Brian Luke offers one PhD project.

We have recently performed genome-wide screens in yeast in order to get a better understanding of how checkpoint adaptation is regulated in response to nutrient starvation. Through this approach we have identified multiple novel genes that appear to have significant effects on checkpoint adaptation. This project would involve working with both yeast as a model system and cultured human cell lines to understand how these genes impinge on checkpoint adaptation and furthermore to characterize how they are related to the nutrient control of DNA damage checkpoints. We will be inducing DNA damage specifically at telomeres, as well as using general chemotherapeutic agents to induce genotoxic stress. Furthermore, we will be anlayzing the effects of checkpoint adaptation in mutations associated with cancer development. Students will employ standard, as well as, high throughput yeast genetics, FACS analysis, cell cycle analysis, DNA repair assays, fluorescence microscopy as well as tissue culture techniques and multiple other molecular techniques in order to monitor checkpoint status.
