PhD position in Molecular Biology - Decoding DNA damage signaling using quantitative mass spectrometry

Diese Stellenanzeige ist abgelaufen. Finden Sie hier zahlreiche ähnliche Jobs.

In the field of  “Decoding DNA damage signaling using quantitative mass spectrometry” the research group of Dr. Petra Beli offers the following PhD project:

Investigation of protein dynamics at DNA lesions

The recruitment of DNA repair factors to the sites of DNA lesions is a complex process that is tightly regulated by posttranslational modifications of proteins such as phosphorylation and ubiquitylation. We have recently combined a DNA-mediated chromatin pull-down technique with quantitative mass spectrometry-based proteomics to investigate the recruitment of proteins to DNA lesions in a highly parallel manner. The prospective PhD student will employ this pull-down technique to investigate the recruitment of proteins and the formation of protein complexes at the sites of specific DNA lesions. These studies will be performed in wild type cells as well as in cells lacking ubiquitin ligases to examine the impact of these enzymes on the protein dynamics at DNA lesions. The mass spectrometric experiments will be complemented by cell biological and genomic methods such as live cell imaging, flow cytometry and next generation sequencing. The prospective PhD student will employ CRISPR/Cas9 genome editing techniques to knockout ubiquitin ligases in human cells. In addition, the student will collaborate with a leading international research group with complementary expertise with an option to perform part of the planned experiments abroad.
