Proteomic analysis of RNAi-pathways in C. elegans


The discovery of RNA interference (RNAi) in 1998 triggered the discovery of many novel, gene-regulatory pathways. Each of these pathways is characterized by small RNA molecules that guide so-called Argonaute proteins to specific targets. At their targets, the Argonaute proteins elicit effects that can range from changes in translation efficiency to RNA de-stabilization and chromatin modification. Most animal species employ multiple RNAi-related pathways, each of them acting in more-or-less specialized gene-regulatory pathways. In general, it is believed that the various proteins acting in the different RNAi-pathways engage different protein-protein interactions in order to bring about the different effects. However, detailed differential interaction patterns of these proteins have not been described. This project will use the nematode C. elegans to address this issue, and aims to develop detailed differential protein-protein interaction maps for the different RNAi pathways that exist in C. elegans. This will be combined with genetic and/or biochemical analysis of the identified factors. The project will involve hands-on experiments dealing with both genetic engineering of C. elegans using novel gene-editing techniques and cutting-edge mass spectrometry, and will be executed in close collaboration between two groups at the IMB, the Ketting and the Butter labs.
