How ribonucleosomes provide global structure to RNA


A particularly interesting protein family to study in the context of pre-mRNA processing and interactive pre-mRNP assembly are the heterogeneous nuclear ribonucleoproteins (hnRNPs). Rivalling histones in their abundance, hnRNP proteins have been described to form hnRNP particles, which have - in analogy to nucleosomes - been referred to as "ribonucleosomes". Their high abundance and presence along most transcripts suggest them as major players in guiding the binding and function of other RBPs. The importance of this protein family is underlined by their involvement in several deleterious neurological disorders such as Amyotrophic Lateral Sclerosis (ALS) and Alzheimer's Disease.
This PhD project will address how hnRNP proteins globally organize pre-mRNAs. The PhD student will combine state-of-the-art genome-wide in vivo RNA biology tools, such as RNA-seq and iCLIP, with genetic and biochemical experiments. This project will contribute to our understanding of the molecular role of hnRNP particles in normal cellular function and disease. The project will rely on close collaborations with computational scientists, and an interest of the PhD student in bioinformatics approaches is beneficial.
