The RADIATE consortium is an EU wide research project to advance our understanding of tumor and tissue responses to ionizing radiation thereby paving the way for a biological optimization of  therapeutic interventions combining radiotherapy with biological targets to improve the outcome of cancer treatment. RADIATE will investigate the following areas: 1) mechanisms of tumor radioresistance and normal tissue toxicity, and 2) the contribution of the microenvironment to the tumor radiation response including the immune changes.

Role of cancer-associated fibroblasts for the radiation-response of solid tumors.
High resistance of human solid tumors to radiotherapy is a major obstacle to successful cancer treatment. Tumor-stroma-interactions play a critical role in the biology of solid human tumors: Moreover, there is increasing evidence that cells from the tumor stroma, particularly endothelial cells and immune cells, affect the efficacy of cytotoxic anticancer therapies. However, the role of stromal fibroblasts for the tumor cell response to radiotherapy remains is still largely unknown. Our work indicates that soluble factors secreted by stromal fibroblasts as well as cell-to-cell contacts between fibroblasts and cancer cells impact on cancer cell proliferation, the activity of growth and survival signaling pathways in the tumor cells, radiation sensitivity and cancer cell survival after radiotherapy. The proposed project aims to clarify the role of fibroblasts in modulating the response of genetically defined cancer cells to radiotherapy, to define the ways in which ionizing radiation modifies the effects of stromal fibroblasts on cancer cells, and to unravel the molecular mechanisms that govern these interactions with a focus on resistance-promoting secreted factors. Newly identified candidate resistance factors that determine fibroblast-mediated radiation resistance will be validated by probing tissues from cancer patients. Novel therapeutic strategies to target fibroblast-mediated radiation resistance will be identified from this work and validated in murine models in vivo. We are looking for highly motivated young researchers with a strong interest in interdisciplinary research projects in cell/tumor biology and Diploma or Master degree in biochemistry, biology or molecular medicine. You will have the opportunity to perform your PhD in an excellent and supporting scientific environment within the framework of the structured PhD program of the EU ITN Radiate. Good knowledge and practical expertise of current cell and molecular biology techniques as well as hands-on experience with radiation biology methods and/or animal experiments is highly desired.
