AIT Austrian Institute of Technology, the Austrian research institute with European format which focuses on the key infrastructure issues of the future, is looking to strengthen its team with the immediate appointment of a
 
PhD Thesis “Development of point of care immunoassays for veterinary saliva diagnostics”
 Saliva has the great potential to enable decentralized point of care (POC) testing performed by laymen such as farmers or breeders in the veterinary field. Although blood tests are established in the central lab and at the veterinarian office, a rapid and userfriendly field-test remains to be developed. In this PhD thesis the whole analysis chain starting from reagent storage, sample preparation, fluidic processing, biosensing and signal acquisition will be implemented in a compact device. The first part includes novel saliva sample preparation methods such as viscosity reduction, filtering and affinity based purification all compatible with point of care diagnostics. The second part includes the development of immunoassays for human and animal saliva based on lateral flow devices which are validated against standard chemiluminescence ELISAs. Assay development will start with CRP, where further analytes will be defined during the project. The third part includes the integration of sample preparation and saliva assay with an electrochemical sensing platform developed by AIT. The assays and purification concepts will be transferred to the electrochemical platform to establish a fully integrated POC device. Finally the device will be validated with human and animal saliva to assess the performance of the device.
 
Job profile:
  • Development of user-friendly saliva sampling and novel sample preparation methods
  • Development of point of care immunoassays (lateral flow and paper fluidic concepts)
  • Validation with standard ELISAs (absorbance and chemiluminescence)
  • Design and implementation of microfluidic reagent storage and process control
  • Evaluation of electrochemical sensing concepts for quantitative saliva measurements and comparison to standard ELISAs
  • Integration of sample preparation, microfluidics and electrochemical sensing into the final point of care device
  • Data analysis, reporting, conferences, publications in peer reviewed journals
     
Required Skills:
  • Study of veterinary medicine, molecular biology, biomedical engineering, process engineering, biotechnology or any related field
  • Interdisciplinary knowledge of biology/medicine and engineering beneficial
  • Motivation for development of novel methods and diagnostic devices
  • Creativity, innovation and strength in unconventional ideas
