PhD student (German salary scheme TV-L E13/2)
 
Our research focuses on molecular mechanisms in motoneuron diseases, especially spinal muscular atrophy with respiratory distress type 1 (SMARD1).
 
The project, financed by the Deutsche Forschungsgemeinschaft aims to:
 
  • Detailed analysis of Ighmbp2 (ribosome-associated ATPase/helicase, affected in SMARD1) and its role in motoneuron and muscle cell degeneration in SMARD1 mouse models.
  • Studies on cultured primary motoneurons from SMARD1 mouse models with focus on defective signaling mechanisms for cellular differentiation and survival under distinct culture conditions.
  • Analysis of functional deficits of Ighmbp2 deficient motoneurons via functional studies e.g. Calcium Imaging and Fluorescence Recovery after Photobleaching (FRAP).
  • Studies in Ighmbp2 deficient muscles and primary myotubes on IGF1 receptor and other growth factor signaling pathways.
  • Functional studies in SMARD1 mice (postnatal) with focus on the contribution of Sodium and Calcium channels to the phenotype.

We are looking for a highly motivated PhD student with knowledge in neurobiology and good technical background in molecular biology (qRT PCR, Northern and Western Blots, cloning and cell culture techniques). The candidate will use advanced molecular biology and cell culture techniques including life cell imaging and iPSC techniques for studying alterations in diseased neurons. Motor function studies, histological analysis of mouse tissue and confocal microscopy will complete the project. The project profits from a strong cooperation and knowledge transfer between partner groups, and is well embedded in the research focus of the institute. He/she will have access to a modern and well-equipped research facility, will work with already available mouse models, and will be supported by technical assistance.
