At the Leibniz ScienceCampus InfectoOptics in Jena infection biologists and photonics scientists work together to develop urgently needed novel optical diagnostic tools for infectious diseases. InfectoOptics combines research in the field of optical high technologies with current topics in infection biology. The InfectoOptics doctoral researchers are integrated into the Jena School for Microbial Communication (, a graduate training program funded by the Excellence Initiative of the German Research Foundation.

Analysis of lipid rafts in fungal model organisms
With this project, the analysis of lipid rafts in the model basidiomycete Schizophyllum commune and the human pathogen Aspergillus fumigatus is combined. For A. fumigatus, a genome-wide knock-out library is being constructed in the working group of Prof. Brakhage which will be available to test potential components of lipid rafts. For the visualization, lipid raft enhanced concentrations of G-protein coupled receptors, flotilllin, but also ergosterol are measured. Since cell wall elongation in growing hyphal tips is a natural place for easy access of signals, a higher signal perception is expected at the hyphal tip. After showing this signaling hotspot in A. fumigatus, this can be verified using the isotropic growth of yeast with C. albicans. The comparison of hyphal versus yeast growth allows a direct correlation of gene functions to filamentous growth by molecular biology and genetic analysis. In addition to fluorescence-in vivo-video microscopy under high resolution conditions, Raman spectrometry and AFM will be used to reveal functional association of lipid rafts with specific signal perception functions and polar hyphal growth.
