BRAIN regularly incorporates life science students into project work within the company. This work always accompanies the completion of a diploma or Master‘s thesis and in some cases it can be part of a practical semester lasting at least 6 months.

“Identification of enzymes for food and/or feed applications by means of sequence- and activity-based screening methods“

Introduction:
Enzymes play a key role in the production of food and beverages. Especially hydrolases, like proteases, amylases, lipases, pectinases, cellulases, and transferases, like glycosidases, are produced in kg- to tons-scale for their application e.g. in the bakery and confectionary industry, in dairy products, cheese and more. Also for the modification of food, for texture improvement, to optimize dough and enhanced shelf-life properties, often enzymes are required.

Project description:
The aim of this project is the identification of enzymes from the BRAIN BioArchive that are able to degrade different substrates and produce valuable product of high interest for the feed and/or feed industry. The project will include the preparation and screening of genome or metagenome libraries (activity-based and sequence-based), the evaluation and comparison of nucleotide/protein sequence data and the characterization of identified enzymes.

We offer the opportunity to extend knowledge in the following methodologies:
  • Microbiology (cultivation of wildtype strains), molecular biology (PCR, cloning) and handling with E. coli
  • Preparation of genome and/or metagenome libraries by state of the art techniques
  • Development of screening assays: activity-based in 96well or 384well plate format (colorimetric, fluorescence)
  • Analysis of expression levels by spectrophotometric or fluorimetric enzyme assays, SDS-PAGE, immunoblots
  • Substrate/product profiling analyses by analytical methods, e.g. gas- and liquid-chromatography
Necessary skills:
  • M.Sc. student in Molecular Biology, Biochemistry, Biotechnology or related studies in Life Sciences
  • Highly motivated and keen to independently push a project
  • Experience in working with sequence databases is required
  • Preparation and evaluation of sequence alignments, design of primer, sequence analysis and sequence assemblies are a must
  • Hands-on experience in the development of screening assays is preferable: activity-based in 96well or 384well plate format (colorimetric, fluorescence)
  • Analytical skills in gas- and liquid-chromatography are helpful
