Description:

We have recently identified a common progenitor to all helper-like ILCs, dubbed CHILP (common helper-like ILC progenitor) (1,2). While clonal differentiation assays have shown that a single CHILP can give rise to all helper-like ILC subsets, the signals determining specification to the various ILC lineages remain unknown. Our preliminary data suggest that the various ILC subsets develop in a timely segregated manner. We will use inducible fate labelling techniques and lineage tracing to determine the ontogenetic windows of ILC development, and then interrogate the signals available at that time to drive ILC lineage specification. In addition, ILC identity seems to be controlled by Id2 but the transcriptional circuitry coordinated by Id2 remains unknown (2,3). We have recently generated mice in which Id2 expression can be extinguished in a time-controlled fashion. This puts us in the position to interrogate the role of Id2 for ILC progenitors and for maintaining ILC identity in mature ILC lineages that continue to express Id2 (3). We further aim to determine the interaction partners of Id2 using mass spectrometry approaches. These studies will clarify the role of Id2 for ILC development, function and maintenance.
