The position will offer the possibility of further scientific qualification in the frame of the as-signed duties.
Scientific services belong to the duties in research and teaching. The candidate will take part in the International Graduate Programme “Molecular Biology and Medicine of the Lung (MBML)” and cooperate within the frame of the LOEWE-Cluster “Medical RNomics”. The emphasis of the pro-ject will be on Long non-coding RNAs in human lung innate immunity. LncRNAs constitute a novel, heterogeneous class of eukaryotic RNAs >200 nt in size, that do not encode proteins. We have recently identified dozens of lncRNA genes that are activated on challenge of human monocytes with bacterial immune agonists. Aim of the current research project is to elucidate the immune-regulatory functions of selected lncRNA in the human innate immune response to bacterial patho-gens of the respiratory system. To this end primary and immortalized human cell cultures will be challenged with the model pathogen Legionella pneumophila and lncRNAs expression changes will be monitored by RNA deep sequencing. Subsequent lncRNA functional characterization will in-volve the identification of interacting protein partners by affinity chromatography as well as over-expression or knockout of lncRNA genes by lentiviral gene transfer and CRISPR/Cas9 technology, followed by mRNA expression profiling. Furthermore, standard techniques in immunology and molecular biology (e.g. ELISA, real-time PCR) will be routinely applied.
Candidates should hold an excellent master or diploma degree in life sciences. A keen interest in current issues of Immunobiology and RNA biology is required as well as extensive practical experience with standard methods of eukaryote molecular biology. Previous experience with cell culture, RNA molecular biology and Infection biology are of advantage. Successful applicants can expect a state-of-the-art laboratory, an innovative research environment and excellent supervision.
