TRON gGmbH – Translational Oncology at the Medical Center of the University Mainz – is a growing biopharmaceutical non-profit organization with the goal to develop highly innovative technologies to address medical needs in the field of cancer diagnostics and therapeutics. TRON is currently expanding its multidisciplinary Clinical Cancer Genomics Unit with exceptional experience in deciphering cancer heterogeneity and effects of immunotherapeutic approaches for the treatment of severe diseases. The team is applying latest single cell RNA-sequencing technologies for gene expression analysis and TCR profiling. In the collaborative framework of the DFG funded Collaborative Research Centre (CRC) “Mechanism of drug sensitivity and resistance in small cell lung cancer (SCLC)”, we are searching two highly motivated PhD students. These PhD students will generate novel insights into T cell recognition and immune editing in SCLC.

In this first position, the PhD student will identify and validate TCRs from interesting T cell phenotypes. Based on these findings a second PhD student “Immune escape in cancer” will study immune editing mediated by T cell recognition in SCLC models. Both PhD students will collaborate in multi-disciplinary teams with other TRON units like Cell Culture, Animal Models, NGS and Bioinformatics to plan, design and run experiments. In addition, they will interact with CRC partners and participate in regular workshops and trainings at University of Cologne. PhD student “Profiling of cancer-reactive T cells (m/f/d)

Activities and responsibilities

  • Establish tumor-infiltrating lymphocyte (TIL) cultures from SCLC tumor biopsies and prepare T cell populations for T cell profiling
  • Perform TCR repertoire analyses of T cell populations via next generation sequencing (NGS)
  • Conduct single cell immune profiling experiments (10x Genomics) and analyse single T cells for their transcriptome and TCR profiles
  • Clone potentially SCLC-specific TCR clonotypes for subsequent specificity testing
  • Perform in vitro co-culture experiments with human T cells transiently transfected with candidate TCRs and SCLC cell lines and subsequent ELISpot assays
  • Develop and evaluate new technologies and protocols for single cell analyses including assays for immune cell isolation, sample preparation, sequencing, and bioinformatics data analysis
  • Plan, implement, document and present research projects and experiments

Qualification profile

  • Master degree in Biochemistry, Cell Biology, Biotechnology or related discipline
  • Deep understanding in molecular biology, cancer biology and immunology
  • Hands-on experience in primary cell culture, cloning and sequencing is required
  • Practical experience in flow cytometry, ELISA and computational data analysis (especially R language) is beneficial
