PhD Position: “Light-activatable tools for the spatio-temporally controlled localization to membranes”

Project Description:
The controlled localization of proteins to specific regions of the cellular membrane is critical to regulate processes involved in signaling, transport, binding interactions, and curvature. Natural mechanisms to recruit proteins to membranes are typically complex and require multiple steps as well as accessory components. Current approaches to specifically target proteins to membranes lack the ability to control the interactions in a spatiotemporal manner.
The project aims to design and develop light-sensitive, lipid-sensing peptides for the on demand recruitment of membrane modulating moieties/ proteins. The spatiotemporal control of interaction will allow for efficient and localized protein attachment to membranes to elicit specific responses such as induced membrane curvature.
The project will consist in the organic synthesis of “caged” peptides, their photophysical characterization and the in vitro analysis of the light-induced binding to the surface of different cell-like compartments. For the characterization of the light-dependent interaction, a broad spectrum of methods and techniques (laser scanning microscopes, SPR, ITC, etc.) is available.

As a member of the interdisciplinary and international research training group CLiC, you will have the opportunity to learn applied organic photochemistry, state-of-the-art imaging techniques as well as cutting-edge photophysical research. We offer a close supervision aiming to form an independent scientist in the field of chemical biology. You will work in an interdisciplinary and forefront-equipped research environment of the CLiC training group, embedded in the Life Science Campus Riedberg, Goethe University.

Your skills and qualifications:
You should have an excellent Master degree (MSc) and a good academic background in organic chemistry, biochemistry, chemical biology or related disciplines. Additional knowledge in solid-phase peptide synthesis and/or cell biology will be appreciated.
